Nach diversen Gerüchten wurde "Resident Evil 6" letztlich Mitte Januar 2012 offiziell von Capcom enthüllt.

Teil 6 umfasst drei parallel verlaufende Handlungsstränge, in denen jeweils zwei Charaktere vorkommen. Die Wege der so insgesamt sechs Protagonisten kreuzen sich immer wieder im Spielverlauf, sodass diverse Situationen aus verschiedenen Blickwinkeln erlebt werden.

Ähnlich dem kommerziell extrem erfolgreichen Vorgänger bietet auch dieser Ableger wieder kooperative Spielmodi. Offline für zwei Spieler im Split-Screen (kein Vollbild, sondern erneut zwei Widescreen-Ausschnitte wie bei Teil 5, sodass am rechten/linken Bildrand schwarze Balken sichtbar sind) oder System-Link, online für vier Teilnehmer.

Sowohl die Kampagne als auch das "The Mercenaries"-Minispiel lassen sich offline und online gemeinsam im Team spielen. Der "Mercenaries"-Modus muss hier nicht mehr freigespielt werden, sondern ist von Beginn an fester Bestandteil des Titels. ABER: In den deutschen Versionen steht dieser Modus wohl erst nach Beenden aller Kampagnen zur Verfügung!
