Die Hintergrundgeschichte von "Resident Evil: The Umbrella Chronicles" befasst sich mit dem Niedergang des Umbrella-Konzerns. Im Verlauf der Handlung tauchen immer wieder bekannte Charaktere auf, unter anderem Jill Valentine, Chris Redfield, Rebecca Chambers, Billy Cohen oder Albert Wesker. Diese bahnen sich ihren Weg durch bekannte Orte und Szenen aus "Resident Evil Zero", "Resident Evil" (Remake) und "Resident Evil 3: Nemesis" sowie mehrere Side Stories, die den Verfall von Raccoon City näher beleuchten. Die Geschichte orientiert sich allerdings nur lose an ihren Vorbildern; vor allem der Part von "Resident Evil 3: Nemesis" wurde stark verändert.
"Resident Evil: The Umbrella Chronicles" ist ein Rail-Shooter, bei dem der Spieler lediglich das Fadenkreuz kontrolliert und sich Gegnerhorden vom Leib halten muss - frei durch die Level bewegen kann er sich nicht.
